چکیده

K. Webbley, S. Harris, J. Bloom, C. Henson, V. Wilson, C. Hawes, D. Price, J. Haffenden Pharmaron, Inc. Purpose Antisense Oligonucleotides (ASO), continue to elicit interest as novel therapeutic agents designed to specifically and selectively inhibit the production of various disease related gene products. For efficacy to occur, the therapeutic material must reach the site of action in quantities sufficient to produce the desired therapeutic effect. An understanding of the pharmacokinetics of ASOs and, in particular, the mechanisms by which oligonucleotides accumulate in tissues and in cells is therefore critical for the development of effective antisense therapeutics.

Restoration of correct splicing in IVSI-110 mutation of β-globin gene with antisense oligonucleotides: implications and applications in functional assay development

Objective(s): The use of antisense oligonucleotides (AOs) to restore normal splicing by blocking the recognition of aberrant splice sites by the spliceosome represents an innovative means of potentially controlling certain inherited disorders affected by aberrant splicing. Selection of the appropriate target site is essential in the success of an AO therapy. In this study, in search for a splic...

Tritium labeling of antisense oligonucleotides by exchange with tritiated water.

We describe a simple, efficient, procedure for labeling oligonucleotides to high specific activity (< 1 x 10(8) cpm/mumol) by hydrogen exchange with tritiated water at the C8 positions of purines in the presence of beta-mercaptoethanol, an effective radical scavenger. Approximately 90% of the starting material is recovered as intact, labeled oligonucleotide. The radiolabeled compounds are stabl...

Specific Inhibition of the Expression of the Promyelocytic Leukemia (PML) Protein by Anti-Sense Oligonucleotides

In the present study, using anti-sense oligonucleotides the inhibition of expression of the PML protein hasbeen investigated. The anti-sense oligonucleotides were designed against the translation initiation site ofthe PML gene, and their effects were investigated on cellular growth and DNA synthesis. Incubation of normalhuman fibroblast cells with the anti-sense oligonucleotid...

Synthesis of 3H-labeled nucleoside-methyl[CT3]phosphonamidite and incorporation into methylphosphonate oligonucleotides for biodistribution and biostability studies.

Antisense oligonucleotides, complementary to certain gene sequences, are designed to hybridize to mRNA and inhibit the synthesis of disease related cellular or viral proteins (1-3). Although numerous in vitro studies have established a great deal of therapeutic potential for these oligonucleotides, there are still many unresolved properties of these oligomers which are critical to drug developm...

Preparation and in-vitro Evaluation of an Antisense-containing Cationic Liposome against Non-small Cell Lung Cancer: a Comparative Preparation Study

The current methods for treatment of cancers are inadequate and more specific methods such as gene therapy are in progress. Among different vehicles, cationic liposomes are frequently used for delivery of genetic material. This investigation aims to prepare and optimize DOTAP cationic liposomes containing an antisense oligonuclotide (AsODN) against protein kinase C alpha in non-small cells lung...
